Abstract

The Docosahexaenoic acid (DHA)-rich lysophosphatidylserine (LPS) with many valuable features was successfully synthesized via enzymatic esterification of DHA and sn-glycero-3-phosphatidylserine (GPS) in vacuum. DHA was prepared and purified from fish oil by urea inclusion method. GPS was prepared from phosphatidylserine (PS) by enzymatic hydrolysis and purified by column chromatography. The enzyme for catalytic reaction was screened, and the Lipozyme TL IM was considered to be more efficient than other lipases and phospholipase in the production of LPS. Under optimized the conditions of temperature 45 °C, enzyme loading 2 g and substrate mass ratio 1:15, the yield of LPS was 88.98 mol%. The results of 31P NMR spectra showed that 1-acyl-sn-glycero-3-lysophosphatidylserine (sn-1-LPS) was the main component of the ultimate product, and the incorporation of DHA was 71.63 mol%.
