Abstract

Electron microscopic autoradiography was carried out on HeLa and Hep 2 cells that were infected with vaccinia virus. Tritiated thymidine was added to the cultures 3 hr after the virus, and the cells were fixed 30–60 min after exposure to tritiated thymidine. Reacted grains occurred over cytoplasmic foci containing fibrillar material that was clearly demarcated from the rest of the cytoplasm but was not bounded by membrane. Most of the foci were devoid of recognizable viral particles and were thought to be cytoplasmic sites of viral DNA synthesis.
