Abstract

Objective Objective to investigate the effects of tanshinone Ⅱ A on proliferation, apoptosis and cell cycle distribution of osteoblasts MC3T3-E1. Methods The osteoblasts MC3T3-E1 were divided into four groups: blank control group, low concentration group (5×10-3 mg/L), medium concentration group (5×10-2 mg/L) and high concentration group (5×10-1 mg/L). The control group was treated with routine cell culture medium without any drugs, while the low concentration group, the middle concentration group and the high concentration group were treated with tanshinone Ⅱ A to culture the cells, methyl thiazol tetrazolium (MTT) assay was used to detect the proliferation of thyroid cancer cells, and flow cytometry was used to detect the cell cycle and apoptosis. Results The proliferation rate of osteoblasts MC3T3-E1 in low, middle and high concentration group was significantly higher than that in blank control group at different time points, and that in high concentration group was significantly higher than that in low concentration group at different time points (F=2.140, 2.130, 3.120, P< 0.05). The apoptosis rate of osteoblasts MC3T3-E1 in low, middle and high concentration groups was significantly lower than that in blank control group at different time points, and that in high concentration group was significantly lower than that in low concentration group at different time points (F=2.390, 2.990, 3.020, P< 0.05). The proportion of cells in G0/G1 phase in low, middle and high concentration group was significantly higher than that in blank control group at different time, and the proportion of cells in S phase and G2/M phase was significantly lower than that in blank control group, andthe proportion of cells in G2/M phase was significantly lower than that in low concentration group and medium concentration group (F=2.310, 2.700, 2.340, 2.170, 4.920, 4.420, 2.840, 2.120, 4.180, P<0.05). Conclusion Tanshinone Ⅱ A can promote the proliferation of osteoblasts MC3T3-E1, inhibit the apoptosis of osteoblasts MC3T3-E1, and significantly improve the cycle distribution of osteoblasts MC3T3-E1.
