Abstract

Objective To investigate the effect of pretreatment of nuclear factor-kappa B (NF-κB) essential modifier binding domain (NBD) on acute necrotizing pancreatitis (ANP).Methods 5% sodium taurocholate was retrograde injected into the biliopancreatic ducts to induce the ANP model.NBD was injected intraperitoneally into the rats 30 minutes before the operation.The rats were sacrificed 6 or 12 h after operation,and the pancreatic histopathology scores,NF-κB P65 protein expression,serum amylase and serum TNF-alpha,IL-6 were detected.Results The serum level of TNF-alpha,IL-6,pancreatic histopathology scores,NF-KB P65 protein positive expression rate were (76±6.7) ng/L,(387±67) ng/L,8.71±0.45 and (49.26±2.15)%,respectively,in ANP group;after NBD pretreatment,the corresponding values were (48±4.5) ng/L,(295±48) ng/L,6.76±0.26 and (37.46±1.68) %,respectively;and the difference was statistically significant (P<0.05).Conclusions The NBD peptides could inhibit the over-sctivation of NFkappa B,down-regulate the expression of IL-6 and TNF-alpha,alleviate the inflammation of ANP.
