Abstract

Objective To explore the effects of morphine on the expression of Survivin and Caspase-9 in human gastric carcinoma MGC-803 cells.Methods MGC-803 cells were divided into two groups.The cells in the control group were incubated with no drugs,while the cells in the morphine group were incubated with 0.1 μmol/L morphine.The apoptosis was assessed by flow cytometry after MGC-803 cells were incubated with morphine for 24 h.The expression of Survivin and Caspase-9 mRNA and protein was detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and Western blotting respectively.Results The apoptosis rate of MGC-803 cells in the morphine group was higher than that in the control group (P < 0.05 ).Survivin mRNA and protein expression was downregulated,while Caspase-9 mRNA and protein expression was upregulated in the morphine group (P<0.05).Conclusion 0.1 μnol/L morphine may promote apoptosis of gastric cancer cells by Survivin downregulation and Caspase-9 upregulation in vitro.
