Abstract

Objective To investigate the effects of low-frequency pulsed electromagnetic fields (PEMFs) onthe expressionand synthesis of paired-box transcription factor7 (Pax7) in the proliferation of C2C12 muscle satellite cells. Methods C2C12 cells were cultured by cell culture. The subcultured cells were divided into the control group, 0.5 mT magnetic field group, 1.0 mT magnetic field group and 1.5 mT magnetic field, and cultured for continuous 5 d. All groups were examined at 1, 2, 3, 4, 5 d, respectively. Immunohistochemical staining was used to determine the changes of cell phenotype. The synthesis and expression levels of Pax7 were determined by PCR, Western-blot. Results Compared with the control group (0.45±0.07) , different intensities of low-frequency PEMFs could induce the Pax7 expression in the proliferation of the in vitro cultured C2C12 muscle satellite cells. Among the four groups, the Pax7 mRNA expression at 4 d in the 0.5 mT group (8.51±0.16) was the highest, and the difference was statistically significant (P<0.05) . Conclusion Under certain conditions, different intensitiesof low-frequency PEMFs can induce the Pax7 mRNA expression in the proliferation of the in vitro cultured C2C12 muscle satellite cells, and further involves in the proliferation and differentiation of muscle satellite cells.
