Abstract
Objective To investigate the effects of nonmitogenic aFGF on the behavioral changes of Parkinson's disease rat model.Methods Seventy-two male SD rats were divided randomly into 4 groups:Shame group,PD group,PD + NS group and PD + aFGF group,and stereotaxic injection of 6-OHDA into the left substantia nigra pars compaeta (SNC) and ventral tegmental area (VTA) of the rats in the latter three groups to develop PD rat model.In the PD + NS group,injected 10 μl NS into the right lateral ventricle of the rats at 2 days interval since 1 day after the injection of 6-OHDA.In the PD +aFGF group,injected 10 μl(0.2 μg/μl) nonmitogenic aFGF into the right lateral ventricle of the rats at 2 days interval since 1 day after the injection of 6-OHDA.In the Shame group,the rats were injected natural saline(NS) instead of 6-OHDA into the left SNC and VTA.Observed and recorded the behavioral changes of the rats at 4 days,1,2,3,4 weeks after the injection of 6-OHDA.Results No rotary behaviors were observed in the Shame group.In PD group and PD + NS group,the start time of PD rats shortened gradually,the continuous time lengthened,and the rotation rate speeded by degrees.The rotary behavior became invariable until 2 weeks after injection.2 weeks later,in PD + aFGF group,the start time [(5.50±1.18)min] was longer than the PD group[(3.60±1.17)min,P<0.05] and the PD+NS group[(3.10±1.02)min,P<0.05] ;the continuous time [(25.90±8.80)min] was shorter than the PD group [(55.40±10.14) min,P<0.05] and the PD + NS group[(53.90±12.27)min,P<0.05] ;the rotation rate [(6.52±1.34)r/min] was slower than the PD group[(12.90±2.21)r/min,P<0.05] and the PD + NS group [(11.80±3.65)r/min,P<0.05].Compared the rotary behaviors between the three groups,the same statistical results as 2 weeks later can be found at 3 or 4 weeks later.Conclusion PD rats have serious behavioral changes,and the nonmitogenic aFGF may better the rotary behavior of the PD rats.
