Abstract

N6-2-O-dibutryl cyclic-3′,5′-AMP and cyclic-3′,5′-AMP but not ATP or AMP brought about epithelial growth, an increase in lipid droplets and granules and an increase in progestin secretion in cultures of granulosa cells harvested from 3–5 mm follicles of 4–6 month old pig ovaries. The effects were similar to those of porcine LH and FSH. Aminophylline potentiated the effect of LH and cyclic-3–,5–-AMP. These data suggest that gonadotrophin induction of luteinization of the granulosa cell may be mediated by an increase of cyclic-3′,5′-AMP due to gonadotrophin action. (Endocrinology 87: 165, 1970) IT HAS BEEN demonstrated that addition of N6-2-O-dibutryl cyclic-3′,5′-AMP (DBC) to cultures of granulosa cells harvested from any sized follicle of ovaries of rhesus monkeys caused the cells to luteinize (1). LH and FSH together produced a similar luteinization response in cells harvested from small 1 mm follicles; LH and FSH alone produced luteinization in medium sized follicles harvested from days 7 to 11 of the m...
