Abstract

5'-Deoxyadenosine inhibits purine ribonucleotide synthesis de novo, as well as synthesis from adenine, guanine and hypoxanthine in Ehrlich ascites tumor cells in vitro. This is due to inhibition of the synthesis of phosphoribosyl pyrophosphate, but phosphoribosyl pyrophosphate synthetase itself is not inhibited. 5'-Deoxyadenosine affects several aspects of glucose metabolism, and it is proposed that this leads, in an as yet unknown way, to a decreased availability of ribose 5-phosphate for phosphoribosyl pyrophosphate synthesis.
