Abstract

Using methyl-α-cyclodextrin (MαCD) mediated outer leaflet phospholipid exchange in cells, we recently showed insulin receptor (IR) activity in CHO cells was supported after substitution replacing plasma membrane outer leaflet lipids with sphingomyelin, which has a propensity to form ordered domains, but inhibited upon substitution with unsaturated phosphatidylcholines (PC), which tend to form disordered domains. We also showed IR activity increased as membrane width increased, as seen when plasma membrane outer leaflet lipids were replaced with saturated PCs with different length acyl chains.
