Abstract

Objective To investigate the effect of lymphocyte immunotherapy (LIT) on the expression level of T-cell immunoglobulin and muein-3 (Tim-3) and galetin-9 (Gal-9) in maternal-fetal interface of spontaneous abortion mice models. Methods The CBA/J×BALB/c normal pregnancy model and CBA/J×DBA/2 spontaneous abortion model were established. Eight normal pregnant mice models without special treatment were included in the normal pregnancy group. Eight normal pregnant mice treated with anesthesia alone were included in the normal pregnancy+anesthesia intervention group. Eight mice models with spontaneous abortion treated with LIT before being caged were included in the spontaneous abortion+LIT intervention group. Eight mice models with spontaneous abortion injected with the same volume of normal saline before being caged were included in the spontaneous abortion+non-intervention group. The placental loss rate in each group was determined during 12-14 d of pregnancy. The protein expression level of Tim-3/Gal-9 in each group was determined by Western blotting, and the mRNA expression level of Tim-3/Gal-9 was determined by quantitative PCR. Results The embryo loss rate in the spontaneous abortion+non-intervention group was significantly higher than that in the spontaneous abortion+intervention group, normal pregnancy group and normal pregnancy+anesthesia intervention group (P 0.05) . Conclusion LIT may effectively decrease the embryo loss rate in spontaneous abortion mice models, and its mechanism may be related to the regulation of Tim-3/Gal-9 expression level.
