Abstract

The effects of CO 2 enrichment (0.3%), light at 60 and 120 μmol m -2 s -1 photosynthetic photon flux density (PPFD) and sucrose concentration in a medium (1-4%) on the in vitro growth, the quality of microcuttings and their subsequent ex vitro rooting and survival in greenhouse were studied. In vitro shoot cultures of rose cv. Sonia and gerbera cv. Rebecca were cultured in vessels allowing ventilation through natural diffusion under the low and high light intensity provided by Warm White fluorescent 36 W lamps. In the case of rose, the CO 2 enrichment had positive effect on ex vitro rooting and establishment of rose shoots only when combined with the lowest level of sucrose in the medium (1%) and high light intensity. Rose shoots were characterised by significantly higher photosynthetic activity compared to those of gerbera. In this genus, the light of low intensity and high sucrose concentration (4%) in the multiplication medium had the best effect on the growth of shoots and their ex vitro rooting. CO 2 enrichment influenced positively the quality of gerbera microcuttings and their rooting at reduced sucrose levels (1-2%) under lower phosynthetic PPFD.
