Abstract
Objective To observe the effects of CC chemokine ligand 5(CCL5)on cell proliferation and the proportion of CD44~+/CD24~-subpopulation in breast cancer cell line McF-7,and to investigate the role of CCL5 in the progression of breast cancer.Methods Effects of recombinant human CCL5(rhCCL5)on cell proliferation of MCF-7 cells were detected by MTT assay,and the percentage of the CD44~+/CD24~-subpopulation in breast cancer cell line MCF-7 was analyzed by flow cytometry.Results When MCF-7 cells were treated with rhCCL5.(10,50,100,500μg/L)for 48 h,the viability of cells was increased in a concentration-dependent manner(P<0.05).The viability of MCF-7 cells treated with 100μg/L rhCCL5 for 48 h was higher than that for 24 and 0 h(P<0.05).The proportion of CD44~+/CD24~-subpopulalion in MCF-7 cell line was elevated to 5.68-fold of the control cells,when treated with 50μg/LrhCCL5(P<0.05),and to 6.05-fold,when treated with 500μg/L rhCCL5(P<0.05).Conclusion CCL5 could elevate cell proliferation capacity and the proportion of CD44~+/CD24~- subpopulation of MCF-7 cells.which might be one of the possible mechanisms of CCL5 stimulating the progression of breast cancer.
