Abstract

Objective To study the effect of 4-N-methylamine-nitroso-1-(3-pyridyl) acetone on the proliferation, apoptosis of lung adenocarcinoma A549 cell. Methods Lung adenocarcinoma A549 cells were treated by medium containing 3% 4-N-methylamine-nitroso-1-(3-pyridyl) acetone. A549 cells cultured for 1 weeks, 2 weeks, 3 weeks were collected separately, and the cell proliferation was detected by MTT method, and the cell apoptosis was observed by Hoechst 33258 fluorescence staining. Results There was no significant difference in the absorbance of A549 cells cultured for 3 weeks in control group (F=0.554, P=0.650). After the 4-N-methylamine-nitroso-1-(3-pyridyl) acetone treated for one week, two weeks and three weeks , the absorbance of A549 cells were 0.516±0.007, 0.485±0.005, 0.432±0.013, the differences were significant(F=171.804, P<0.001). There were significant differences in the absorbance between each treatment group and control group(P all<0.001). With the acting time of 4-N-methylamine-nitroso-1-(3-pyridyl) acetone on A549 cells increased, the proliferation inhibition rate gradually increased, respectively was (22.28±1.58)%, (27.06±0.70)%, (34.60±2.18)%, the difference were significant (F=169.77, P<0.001). 4-N-methylamine-nitroso-1-(3-pyridyl) acetone induced apoptosis of A549 cells, with the extend of acting time, the apoptosis rate in each treatment group gradually increased, which were (16.32±0.93)%, (22.92±1.11)% and (29.88±0.81) %, compared with the control group [(10.22±1.29)%], the differences were significant (F=387.171, P<0.001). Conclusions 4-N-methylamine-nitroso-1-(3-pyridyl) acetone can inhibit proliferation and promote the apoptosis of A549 cells.
