Abstract
We produced the human kidney Man 9-mannosidase in Escherichia coli and studied the effect of the α-1,2-mannosidic linkage located in the α branch of Manα1–6(Manα1-2Manα1-3)Manα1-6(Manα1-3)Manβ1-4GlcNAcβ1-4-GlcNAc, an N-linked oligosaccharide, on the enzyme activity. The α1,2-mannose residue influenced the rate of hydrolysis of and substrate preference for other α-1,2-mannosidic linkages.
