Abstract

CRISPR-Cas effector complexes became widely used in a broad range of genomeengineering applications. Due to simplified cellular delivery, miniature effectorcomplexes such as Cas12f1 with only ∼400-500 aa became of interest and theirgenome editing activity was only recently demonstrated. On the mechanistic level, theprocesses underlying target recognition and DNA cleavage have, however, not beenresolved.Here, we obtain mechanistic insight for Cas12f1 from Syntrophomonas palmitatica.Using mass photometry we characterized the complex composition of the Cas12f1-RNA-complexes.
