Abstract

Due to constant outbreak of deadly microbial pathogens and threats of bio-warfare or -terrorism, developing of novel diagnostic device is highly necessary. In spite of the high selectivity, high accuracy and high sensitivity of conventional biosensors, they could not meet the situations that require the changing of analytical target. Thus programmable biosensor is proposed here to flexibly change the target analyte by changing sensing probe molecule on-site and on-demand. We propose to synthesize short peptide as probe molecule on-chip following traditional Fmoc-solid phase peptide synthesis strategy. Linear hepta-peptide probe (-NH-PPGQPHH-NH2) is synthesised on-chip (optimized conditions of 15 min coupling time and 5 µl/min of flow rate) and successfully performed in situ detection of the target biomolecule (streptavidin) on the same microchip.
