Abstract

A new method for the determination of Hb-binding capacity of serum is described. Serum previously incubated with Hb, during paper electrophoresis at pH 7.0 is separated into three fractions containing free Hb, Hp-bound Hb and MetHb respectively. Concentrations of individual protein fractions are determined by the tannin micromethod. The concentration of the bound Hb is calculated by subtracting the protein content of the serum fraction containing, inter alia, Hp, from that containing the Hp-Hb complex. By the described method the Hb-binding capacity of 20 normal sera was determined. Obtained values were in accord with those reported in the literature.
