Abstract

Chicken egg yolk immunoglobulin (IgY) is the only antibody existing in chicken egg yolk, which has many significant advantages. We developed an immunity affinity biosensor system based on resonance Rayleigh scattering (RRS) and using immunity affinity rabbit anti-chicken IgY (anti-IgY) labeling Au nanoparticles (AuNPs). This system was rapid and sensitive for IgY detection due to the IgY-anti-IgY affinitive immunity interaction. The RRS and absorption spectra of AuNPs and the binding system were analyzed. In addition, the effects of several factors on the scattering intensity were investigated. Under optimal conditions, RRS intensity of anti-IgY-AuNPs increase linearly with an increasing concentration of IgY from 4 to 200 ng/mL. The limit of detection was 1.92 ng/mL. This rapid and selective IgY detection method was used in synthetic samples and egg products with recoveries between 89.86 and 110.25 %, thereby demonstrating the feasible and practical application of this assay.
