Abstract
Desmosomes are macromolecular cell-cell junctions critical for maintaining adhesion and resisting mechanical stress in the heart and epithelial tissue. Dysregulation of desmosomal proteins and desmosome function occurs in genetic and acquired diseases of the heart and skin and is implicated in cancer progression, highlighting the important role these junctions play. Desmosomes have a complex architecture, containing transmembrane cadherins and cytosolic plaque proteins. Desmoplakin is an essential desmosome protein which links the desmosomal plaque to the intermediate filament cytoskeleton.
