Deoxyribose Phosphate Aldolase from Rat Liver

  • The ratio of deoxyribose to total phosphorus was 1; the product was found to have reducing power when determined under conditions which would protect against the hydrolysis of deoxyribose-1-P [9]; paper chromatography in three different solvents with subsequent identification of sugar phosphates by the method of Bandurski and Rxelrod [10] and deoxyribose by the vanillin-perchloric acid reagent of Ma&Alan, Randall, and Smith [11] gave identical RF values with authentic deoxyribose-5-P; and acetaldehyde and glyceraldehyde-3-P were shown to be the-products of the reversible cleavage of the reaction product when assayed in the presence of deoxyribose-5-P aldolase, alcohol dehydrogenase, and glyceraldehyde-3-P dehydrogenase by the method suggested by Racker [1]

  • The liver enzyme apparently requires a carboxylic acid for maximal activity, a finding similar to that in the case of deoxyribose phosphate aldolase purified from L. plantarum [2] and the liver preparation catalyzing the cleavage of deoxyribose-5-P [4]

PROCEDURE

Materials-In all cases, commercial preparations were used unless otherwise indicated. 2-Deoxyribose-5-P was prepared. Methods-In the case of all enzymatic studies, formation of deoxyribose-5-P was measured calorimetrically by the procedure of Dische [5] as modified by Burton [6]. For the determination of deoxyribose-5-P in the isolated product, the method of Dische [5] was used. Organic phosphorus was measured as the inorganic phosphorus formed after digestion with 10 N H2S04. Glyceraldehyde-3-P was measured spectrophotometrically by the increase in absorbance at 340 ml.c in the presence of glyceraldehyde-3-P dehydrogenase and DPN. Deoxyribose phosphate aldolase activity was measured by the amount of deoxyribose-5-P formed from acetaldehyde and glyceraldehyde-3-P generated from fructose 1,6-diphosphate and crystalline muscle aldolase
