Darwish et al. report the preparation of two nanoformulations to enhance delivery of RH to the brain: a self-assembled core-shell nanocapsule of RH and methoxy poly(ethylene glycol)-b-poly(caprolactone) coated with chitosan, and a RH-loaded chitosan-alginate polyelectrolyte nanocomplex. The mucoadhesive properties endowed by the chitosan and alginate promote RH delivery across the nasal mucosa and higher brain uptake of RH is seen in albino mice when the formulations are administered intranasally compared with intravenously.
