Abstract
15557 Background: Approximately 90% of PC harbor mutations in k-ras codon 12. Smoking (Sm) is the most important risk factor for development of PC. Ras genes are critical targets for chemical carcinogens and a relationship between tobacco carcinogens and codon 12 k- ras mutations has been suggested. In lung adenocarcinoma, k-ras mutations have been identified preferentially in patients with a positive Sm history. In PC, a correlation of Sm history with specific k-ras mutations remains to be defined. Our aim was to evaluate whether a correlation between Sm history and k-ras mutation in pts with PC exists. We specifically looked at the correlation between Sm history and aspartic acid (D), valine (V), arginine (R), and serine (S) k-ras mutations. Methods: We retrospectively obtained the Sm history in a cohort of pts enrolled on a prospective pilot trial of k-ras vaccination in resected PC. Genomic DNA samples were analyzed for codon 12 k-ras mutations using multiplex competitive oligonucleotide probe ligatio...
