Abstract
A rotating enzyme-immobilized reactor and a flat pH electrode have been incorporated into a sealed cell for use under continuous-flow/stopped-flow operation. The system has been used for rapid determination of penicillins G and V in pharmaceutical tablets and injectables. Detections based on the increase of hydrogen ion concentration due to enzymatic hydrolysis of the penicillins by the enzyme penicillinase [EC 3.5.2.6] immobilized on controlled-pore glass. Selective determination of the analyte in the 0.001–0.50 mM concentration range can be accomplished at the rate of 30 samples h −1.
