Abstract

Objective To investigate the feasibility of endothelial progenitor cells (EPCs) promoting angiogenesis of tissue-engineered sheet graft using bladder epithelial cells and smooth muscle cells seeded on the bladder acellular matrix (BAM) in vivo.Methods BAM of rabbits was made in vitro.Bladder epithelial cells,smooth muscle cells and EPCs of rabbits were cultured and expanded in vitro,then seeded on the BAM and cultured for 7 days for producing sheet graft.The experimental group was bladder epithelial cells-smooth muscle cells-EPCs-BAM sheet graft and the control group was bladder epithelial cellssmooth muscle cells-BAM sheet graft.Then the grafts were implanted into the back of rabbits.Specimens were obtained at 8th week postoperatively and were examined histologically.Results Bladder epithelial cells,smooth muscle cells and EPCs were cultured and expanded in vitro.At 8th week after operation,histological detection of specimens showed angiogenesis and epithelial coverage in the experimental group.The epithelial cells gradually integrated with the BAM and smooth muscle cells grew well.In the control group,the graft showed contracture and discontinuous epithelial lay,and there was no obvious vascularization.Conclusion Tissue-engineered sheet graft constructed by EPCs,bladder epithelial cells and smooth muscle cells grows well in vivo.EPCs can promote angiogenesis of tissue-engineered sheet graft.
