Abstract

Objective To construct a recombinant adenoviral vector carrying the human BMP-2 gene which may be applicable in gene therapy.Methods BMP-2 gene was first cloned into adenoviral shuttle plasmid to generate recombinant plasmid pAdCMV-BMP-2,which was subsequently co-transfected into 293 cells with pJM17 using LipofectAMINE,and adenoviral vector Ad-BMP-2 was produced by in vivo recombination.Results Insertion of gene and the recombination of adenovirus were identified by restrict enzyme digestions and PCR.Conclusions Construction of adenoviral vectors were successfully achieved and may be available in the future study of BMP-2 gene therapy.
