Abstract

Objective To explore the growth inhibition effect of dihydroartemisinin (DHA) combined with Cisplatin (Cis) and the expression of Caspase-6 and Lamin A/C to human lung adenocarcinoma H1299 cells. Methods The cells were cultured in vitro and divided into four groups: DHA group, Cis group, DHA plus Cis group and control group.Cell inhibition rate were evaluated by thiazolyl blue tetrazolium bromide assays.Flow cytometry was used to detect the apoptosis induced by different treatment groups.Nuclear staining with Hoechst 33258 showed morphology of the nucleus treated with different cultivation methods.The apoptotic-associated proteins Caspase-6 and Lamin A/C were assessed by Western blot. Results Inhibition rate of H1299 cell line by DHA and Cis treated in combination was significantly higher than that in single drug groups.Combination of DHA and Cis had synergistic inhibitory effect on H1299 cell line.Combination of DHA and Cis had pro-apoptotic effect on H1299 cell line.The expressions of cleaved-Caspase-6 and cleaved-Lamin A/C by DHA and Cis treated in combination were higher than those in other groups. Conclusions This study revealed the inhibition of the H1299 cells in the DHA plus Cis group was significantly higher than that in the single treatment group by increasing the expression of cleaved-Caspase-6 and cleaved-Lamin A/C.
