Abstract
Fluorescent tags are usually distinguished by their color, making fluorescence multiplexing at the single-molecule level challenging since only a few tags can be distinguished due to the overlap in their emission spectra and the low signal-to-noise ratios inherent in single-molecule measurements. We have engineered a series of spectroscopically distinguishable single-molecule labels whose emission is characterized by unique combinations of photophysical parameters including brightness, lifetime, and emission spectra.
