Abstract

Objective To investigate the effects of4,5,6,7-tetrabromobenzotriazole (TBB),which inhibits CK2,on human breast cancer cell line MCF-7.Methods The effects of TBB treatment on MCF-7 cells were examined by using methyl thiazol tetrazolium (MTT) assay.The early apoptosis rate and cell cycles were measured by flow cytometry.The mRNA levels of Caspase-3,Caspase-8 and bcl-2 were detected by using Real-time polymerase chain reaction (PCR).The nuclear factor-κB (NF-κB) activities were analyzed by electrophoretic mobility shift assay (EMSA).Results The inhibition rate of MCF-7 cells was increased with the increase in concentration of TBB.After MCF-7 cells were treated with TBB ( 100 p,mol/L) for 24 h,the early apoptosis rate was increased to ( 15.10 ± 0.65 ) % and the proportion of MCF-7 cells in at G0/G1 phase was increased to 76.78%.Besides,the mRNA levels of Caspase-3 and Caspase-8 were 3.410 and 5.736 times higher,while the mRNA levels of Bcl-2 and the activities of NF-κB were decreased (P < 0.05 ).Conclusion CK2 inhibitor TBB promotes the apoptosis of MCF-7 cells,which may be concerned with the reduction of NF-κB activities.
