Abstract
The reconstitution of native tonoplast vesicles from Commelina communis guard cell protoplasts into cholesterol-free planar lipid bilayers revealed single ion channel activity. An 8.1 pS ion channel was found to have a low selectivity for cations over anions (P K :P CI = 1:0.83) in asymmetrical conditions (100 cis:10 trans mM KCI). Addition of cholesterol (50 mol%) to the bilayer induced instantaneous and reversible closure of the channel.
