Abstract

Atomic force microscopy (AFM) is a powerful tool for investigating DNA structure and DNA-protein interactions. In this work, we characterize the assembly and disruption of protein-nucleic acid complexes using atomic force microscopy in liquid. We study interactions within several systems, including eukaryotic nuclear DNA binding protein and human innate immunity proteins. To do this, we developed protocols (AP-mica) for imaging DNA, DNA-protein complexes, and nucleosomes and nucleosome-protein complexes in liquid.
