Abstract
The changes of 3H-yohimoine and 3H-clonidine binding sites in rat vas deferens on treatments with 2-chloroadenosine(2CA) and reserpine were examined with binding experiments. The results demonstrated that treatment with 2CA increased 3H-clonidine binding sites, whereas the treatment did not have effects on 3H-yohimbine binding sites. On the other hand, reserpine treatment increased both 3H-clonidine and 3H-yohimbine Dinding sites and also decreased the IC50 values of Clonidine on 3H-yohimbine binding sites. GTP and islet-activating protein inhibited the effects of 2CA on 3H-clonidine binding. The results imply that treatment of 2CA induced a qualitative change of alpha-2 adrenoceptors, while treatment with reserpine caused Doth qualitative and quantitative changes of the receptors. It is suggested that the qualitative change was mediated through a GTP-induced procès s.
