Abstract
The aim of this chapter is to provide insights into stimulus-responsive liposomes as smart nanocarriers for drug delivery applications. An overview of the main categories of stimulus-responsive liposomes, their characteristics and their added value for drug delivery, controlled release and targeting is presented. The field of stimulus-responsive liposomes encompasses several different preparations that can be led to degradation after the application of an external or internal stimulus. They can encapsulate a gamut of active pharmaceutical ingredients, especially the lipophilic ones, providing low nanotoxicity and high viability for healthy cells, leading to increased bioavailability in damaged tissues. pH-, thermo-, light, ion, enzyme and magneto-responsive liposomes are discussed in depth regarding their characteristics and applications. Special attention is given to the dual- or multiresponsive liposomes, which are used for theragnostic applications. Examples from recently published studies are cited. A comprehensive update about their application in different diseases as well as potential challenges for future drug development are also discussed.
