Abstract

Systems metabolic engineering, which integrated traditional metabolic engineering with systems biology, synthetic biology, and evolutionary engineering, has recently emerged and been used to facilitate construction of high-performance strains. At present, many tools and strategies of system metabolic engineering are developed for cell factories construction and chemicals biosynthesis. In this chapter, general strategies of systems metabolic engineering are reviewed, focusing on developments in system-level metabolism analysis for metabolic model construction, design and construction of novel biological parts/systems, metabolic pathway reconstruction and optimization, and evolutionary strategies assisted improved cell factories performance. Finally, the challenges and perspective for the systems metabolic engineering with functional expansion and application were discussed as well.
