Abstract
This chapter gives an overview of the underlying difficulties in translating data into clinically actionable knowledge. The state of data-centric systems and computational/systems biology methods is described as an alternative to reductionist, molecule-, pathway-, and physiologic endpoint-centric conceptual frameworks. The overarching concepts of data-driven and dynamic mechanistic modeling are introduced, as are general strategies for their use.
