Abstract

Heterocycles are considered the most useful core in drug design and discovery. Pyrimidine ring containing fused five-membered structure showed important pharmacological interventions. A Pinner condensation reaction can easily do the construction of pyrimidine scaffolds. These scaffolds were also found to improve druggability and pharmacokinetic profile. Many naturally occurring drug and drug-like molecules also possess a fused five-membered pyrimidine ring as a core structure, for example, thiamine, riboflavin, folic acid, etc. This chapter discusses the synthetic and applicability aspects of the five-membered fused pyrimidines derivative from 2015 to till date; also, we covered the recently filed patents containing this scaffold.
