Abstract
Method development includes all those steps involved in the selection and optimization of the separation system for each sample. An extensive discussion of solvent strength and selectivity as key features for mobile phase selection and optimization is provided. Optimization strategies are described for normal-phase separations on silica gel and polar chemically bonded layers and for reversed-phase separations on chemically bonded layers. Sample properties are linked to mode selection to allow an informed choice of initial experiments. Gradient methods are described for the separation of mixtures with a wide range of retention properties.
