Abstract

The present chapter outlines in brief the basics of fluorescence spectroscopy explaining the principles, instrumentation, and fundamental concepts of intrinsic and extrinsic fluorescence. The technique is important for understanding the secondary and tertiary structural changes in macromolecules, especially proteins. It also provides deep insights related to protein stability, protein–protein interaction studies with the help of suitable examples. The basic concepts and equations underlying fluorescence polarization and fluorescence resonance energy transfer are highlighted using mathematical equations.
