Abstract

The mammalian target of rapamycin (mTOR) is a central regulator of cellular and organismal growth and homeostasis. In response to various environmental stimuli, mTOR regulates major cellular metabolic processes, including macromolecular synthesis, ribosome biogenesis, and autophagy. In metabolic organs, mTOR mediates whole-body energy balance. Deregulation of mTOR signaling is implicated in various metabolic and endocrine diseases, including obesity, insulin resistance, diabetes, and dyslipidemias. Here we discuss mTOR regulation and signaling in modulation of cellular and organismal metabolism in the context of major metabolic and endocrine disorders.
