Abstract

Commercial multiplex kits capable of amplifying multiple DNA loci in a single tube reaction are essential to solving cases involving DNA evidence in crime labs. In a previous experiment, a selected commerical multiplex kit was used to amplify previously extracted DNA from buccal swabs and simulated evidence samples using the polymerase chain reaction following the manufacturer’s instructions. In this experiment, the allele profile of the amplified loci for the DNA-containing samples will be determined by separating the amplicons using capillary electrophoresis using a single or multiple capillary instrument. GeneMapperID or similar software will be used to make the allele calls and all calls will be verified. The amplified loci and allele ladder will be determined by the kit employed. The allele calls will be tabulated and reported for locus and each sample run.
