Event Abstract Back to Event Changes of cortical electrical activity and open field motility in WAG rats during several weeks of oral manganese exposure Szabolcs Takács1* and András Papp1 1 Szeged University, Medical School, Department of Public Health Medicine, Hungary Long time exposure to manganese causes well known neurological disorders in humans, manifested mostly in a Parkinson-like syndrome with motor dysfunctions, mood and memory problems etc. Electrophysiological alterations following Mn exposure, including epileptic activity, were also reported. These effects have been successfully modelled in animals but data on simultaneous changes at different levels of central nervous activity, electrophysiology and motor behaviour, have been scarce. In the present work, rats were equipped with chronically implanted epidural electrodes and a connector crown for leading off electrocorticogram (ECoG). Adult male (ca. 300 g) animals of the WAG/RijCrl (WAG) strain were used, a strain with inherited tendency to petit mal type epileptic activity. After operation and 10-13 days of healing, the rats had a 60-min session in an open field (OF) box once a week during which ECoG was also recorded. Following 2 pre-treatment control weeks, 4 of the 6 rats received oral Mn exposure via the drinking water (7.5 mg Mn/ml), and the ECoG+OF recording was carried out for further 6 weeks. In the treated rats there was a gradually developing decrease in the ECoG power, predominantly above 30 Hz. A time-dependent change in the ECoG spectrum was present both in the treated and control animals but was dissimilar. The OF motility decreased in every animal (an effect of age). Irrespective of that, the treated rats had longer periods of immobility in the first 2 weeks of treatment, and transient increase of the ambulation distance in the next week. In the observed behavioural and electrophysiological changes, both the own effects of Mn and its interaction with the epilepsy-prone baseline of the WAG rats had most likely a role.
