Abstract
Objective To investigate the change of distribution of non-adrenergic non-cholinergic nerve and neuropeptides in the lung of asthmatic rat.Methods Twenty Wistar rats were divided into asthma group and control group.Immunohistochemistry and computer image analysis were used to observe the change of the calcitonin gene-related peptide(CGRP)and vasoactive intestinal peptide(VIP)immunoreactive (IR)nerve fibers.The contents of CGRP and VIP in lung tissues were detected by ELISA.Results ①There were significant changes in distribution and density of CGRP-IR and VIP-IR nerve fibers in airways of asthma group.The relative positive staining areas and mean optical density(MOD)of CGRP-IR nerve fibers increased significantly in asthma group compared with those of control group(P<0.01).But the relative positive staining areas and MOD of VIP-IR nerve fibers decreased significantly in asthma group compared with those of control group(P<0.05 or P<0.01).②The content of CGRP in the BALF of asthmatic rats was significantly higher than that in control group.There was a significantly positive correlation between CGRP and the relative positive staining areas(r=0.903)and MOD(r=0.880)of CGRP-IR nerve fibers.The contents of VIP in the BALF of asthmatic rats were significantly lower than those in control group.There was a significantly positive correlation between VIP and the relative positive staining areas(r=0.899)and MOD(r=0.878)of VIP-IR nerve fibers.Conclusions The distribution of non-adrenergic non-cholinergic nerve were abnormal in the lung of asthmatic rats,which might immediately cause the change of neuropeptides.
