Abstract

Analytical subcellular fractionation and histochemistry were used to investigate the localisation of mannitol oxidase activity in the digestive gland of four species of terrestrial gastropod. In all four species mannitol oxidase was associated with particles that banded at approximately 1.15 g/ml in isopycnic density gradients. In three species mannitol oxidase was associated with particles of intermediate sedimentation rate, while in Limax flavus fast and slow sedimenting particles with mannitol oxidase were observed. Examination of fractions enriched in mannitol oxidase activity indicated similar properties for the enzyme in the three genera investigated. In cryostat sections processed to localise mannitol oxidase large punctate dye deposits were distributed in a characteristic pattern throughout the digestive gland. Our results indicate the presence of distinct organelles carrying mannitol oxidase activity in terrestrial gastropod digestive gland.
