Abstract

This chapter discusses an experiment in which canine littermates were studied to determine the relationship between lymphocyte defined (LD) and serologically defined (SD) antigens and the cell-mediated lympholysis (CML) test in the dog. In the experiment, mixed leukocyte culture (MLC) were set up using an equal number of responding and irradiated stimulating cells. PHA stimulated 51 C r labeled lymphocytes were used as targets. On day 7, the CML test was performed using an effector/target cell ratio that ranged between 80–100:1.In most littermate pair studies, mixed leukocyte cultures (MLC) activation and an SD difference were required for specific cytotoxicity. The cytotoxicity in the absence of a detectable SD difference or in the absence of demonstrable MLC activation suggested that additional determinants were involved in the CML reaction in dogs.
