Abstract
Caveolins are a member of membrane-bound scaffolding proteins which could negatively regulate and compartmentalize signaling in caveloae. Loss of caveolin-1 (Cav-1) expression occurs in the pathogenesis of tumors, and loss of Cav-1 expression in cancer associated fibroblasts contributes to the activation of tumor microenvironment, thus advancing early recurrence and metastasis in prostate and breast cancer. Recent studies have indicated loss of stromal Cav-1 expression is a predictor of clinical outcome in cancers. Finally, we discuss the mechanism of loss of stromal Cav-1 facilitating the cancer progression, including extracellular matrix remodeling, fibrosis and origin of cancer associated fibroblasts.
