Abstract

Bone remodelling consists in cycles of bone resorption and formation executed mainly by osteoclasts and osteoblasts. The healthy functioning of bone remodelling is disrupted by diseases such as Multiple Myeloma and bone metastatic diseases. In this work, a model with differential equations is proposed, including PK/PD of several drugs, and accounting for drug resistance in two variations of the model, corresponding to different paradigms as to the origin and development of resistance. Optimisation of drug dosage is done with Nonlinear Model Predictive Control, a democratic variant of Particle Swarm Optimization, and a fractional order objective function. Obtained prescriptions provide useful conclusions on dosage strategies and resistant cancer behaviour.
