Abstract

Fungal indole diterpenes (IDTs) are isolated as mycotoxins and antiinsectants. Their characteristic molecular structures have prompted research on the mechanisms of IDT biosynthesis. Recent technology in DNA sequencing and bioinformatics has enabled rapid access to the biosynthetic gene clusters of IDTs. Based on cluster information, various approaches provide sufficient data to discuss the IDT pathway. In particular, the reconstitution of biosynthetic genes in a heterologous host is employed to elucidate their functions. This powerful and versatile method not only enables producing final IDTs but also provides individual intermediates. Through analysis of eight representative IDT biosyntheses, we describe herein a detailed mechanism of IDT core construction and further modifications for generating highly elaborated IDTs.
