Abstract

Preparation of calibration curve is one of the main sources of measurement uncertainty for quantitative determinations. A LC-MS/MS method for the quantification of urinary methylphenidate and retalinic acid has been evaluated by observing concentration-response curves, linearity and goodness of fit analysis. The calibration functions were calculated and fitted by linear and quadratic regression models with three different weighting factors. Also the correlation coefficient, the residuals plot and F-test were examined for the linearity of calibration curve. In this study, weighted (1/x²) linear regression was found to give more accurate results and was therefore selected.
