Abstract

A new glucose analyzer is described in which an enzyme reaction coupled with a redox process is monitored amperometrically. The sensor consists of a platinum electrode coated with a thin enzyme (glucose oxidase) layer, separated from the test solution by a semipermeable membrane. The instrument allows the rapid determination of glucose (response time, 40 to 50 sec) by means of a semiautomated procedure. Optimal operational conditions with respect to linearity, stability, and response time are studied and discussed. The influence of different sample characteristics on the measurement (pH value, protein content, medium composition, concentration range, cell or particle concentration, etc.) are investigated and optimized to allow glucose determination in microbiological media.
