Abstract

The article discusses the development of a new method of producing Laetiporus sulphureus (Bull.) Murrill basidiomycete mycelium immobilized on a bacterial cellulose matrix. Mycelium contains biologically active compounds with antimicrobial activity against Gram-positive bacteria, including resistant staphylococcus. The aim of the work was to obtain immobilized mycelium by co-cultivation of L.sulphureus with the producer of bacterial cellulose Gluconacetobacter hansenii. The authors found that when co-culturing the basidial L.sulphureus strain with the bacterial cellulose producing G.hansenii strain, productivity increases by 3.2 times on H5/1 synthetic medium and by 1.9 times on natural Maltax-10 medium (concentration 5%). The resulting immobilized L.sulphureus mycelium has antibacterial properties; its aqueous extracts contain glucans.

Ïîëó÷åíèå ìèöåëèÿ è ìàòðèö äëÿ èììîáèëèçàöèè ïðîâîäèëè ñ èñïîëüçîâàíèåì øòàììîâ: L.sulphureus (Bull.) Murrill øòàìì MZ-22 (ÂÊÌ F-4276D) — êñèëîòðîôíîãî áàçèäèîìèöåòà è G.hansenii GH-1/2008 (ÂÊÏÌ Â-1054) — ïðîäóöåíòà áàêòåðèàëüíîé öåëëþëîçû [10, 14]. Èììîáèëèçàöèþ ìèöåëèÿ ïðîâîäèëè ïðè ñîâìåñòíîì êóëüòèâèðîâàíèè áàçèäèàëüíîãî ãðèáà L.sulphureus ñî øòàììîì-ïðîäóöåíòîì áàêòåðèàëüíîé öåëëþëîçû G.hansenii â äâóõ âàðèàíòàõ: íà ñèíòåòè÷åñêîé ñðåäå ñîñòàâà, ã/ë: ãëþêîçà — 7,0; äðîææåâîé ýêñòðàêò — 5,0; Na2HPO4 — 0,27; K2HPO4 — 0,2; (NH4)2SO4 — 0,3; ìîíîãèäðàò ëèìîííîé êèñëîòû — 0,115, ðÍ ñðåäû 6,5 è íà íàòóðàëüíîé ñðåäå Ìàëüòàêñ-10 (5%). Êîëè÷åñòâåííîå ñîäåðæàíèå ãëþêàíîâ îïðåäåëÿëè â ìèöåëèè áàçèäèîìèöåòîâ L.sulphureus, ïîëó÷åííîì êàê ïðè ìîíî-, òàê è ïðè ñîâìåñòíîì êóëüòèâèðîâàíèè ñî øòàììîìïðîäóöåíòîì áàêòåðèàëüíîé öåëëþëîçû G.hansenii. Êîëè÷åñòâî êàðîòèíîèäíûõ ïèãìåíòîâ, ñîäåðæàùèõñÿ â ñïèðòîâûõ ýêñòðàêòàõ ìèöåëèÿ øòàììà L.sulphureus, ïîëó÷åííîì, êàê ïðè ìîíî-, òàê è ïðè ñîâìåñòíîì êóëüòèâèðîâàíèè ñ G.hansenii îïðåäåëÿëè ôîòîìåòðè÷åñêèì ìåòîäîì ïðè äëèíå âîëíû 450 íì. Äëÿ ýêñïåðèìåíòîâ ïî îïðåäåëåíèþ àíòèìèêðîáíîé àêòèâíîñòè òåñòèðóåìûõ ñîåäèíåíèé ãîòîâèëè èíîêóëþì áàêòåðèàëüíûõ òåñò-êóëüòóð, äëÿ ÷åãî èñïîëüçîâàëè ÷èñòóþ ñóòî÷íóþ êóëüòóðó áàêòåðèé, âûðàùåííûõ íà ñîîòâåòñòâóþùèõ ïëîòíûõ ïèòàòåëüíûõ ñðåäàõ ïðè òåìïåðàòóðå 35°C. Ïëîòíîñòü áàêòåðèàëüíûõ ñóñïåíçèé ñîñòàâëÿëà 0,5 ïî ñòàíäàðòó ÌàêÔàðëàíäà (1,5 108 ÊÎÅ/ìë) è ïðè äàëüíåéøåì ðàçâåäåíèè áóëüîíîì Ìþëëåðà–Õèíòîí â 100 ðàç êîíöåíòðàöèÿ ìèêðîáíûõ êëåòîê ñíèæàëàñü äî 5 105 ÊÎÅ/ìë
